These results indicate that Scalloped and Yorkie alter each other’s behaviour within the nucleus and give insights into how the Hippo pathway regulates gene expression. 7/8.

We wondered whether transcriptional coregulators might change Scalloped’s behaviour at these sites, as a mechanism to alter gene expression. We found using FRAP that the Yorkie activator increased Scalloped’s binding time, whilst repressors decreased long binding events. 6/8.

By imaging single Scalloped molecules, we found their binding duration was extended at these sites relative to elsewhere in the nucleus. 5/8.

We noticed regions in the genome where the transcription factor Scalloped and its coactivator Yorkie were both highly enriched, which were high in markers of transcriptionally active chromatin. 4/8

We saw that Yorkie explored the nuclear environment more rapidly than Scalloped, but Yorkie’s behaviour was highly dependent on the levels of Scalloped within the nucleus. 3/8.

We thought the easiest way to investigate these processes was to watch them, so imaged the giant nuclei in salivary glands of D. melanogaster larvae. This enabled visualisation of transcription factor movement throughout the nucleus as well as DNA binding behaviour. 2/8
