What started as a crazy idea (and therefore a grad student rotation project), has now evolved into a mature body of work, spearheaded by @ChristinaRamel1 . So many potential applications of this approach!.

SPARO works in vivo! We applied this to neurons and astrocytes in adult mouse brain, and used both “omes” to assess protein:RNA concordance.

We leveraged cytosolic TurboID to label RNA binding and Ribosomal proteins, while labeling other cytosolic proteins. We enriched biotinylated proteins for proteomics, and amazingly enough, a whole bunch of RNA came along for the ride… for RNA ‘omics’.
