Interesting for #Cilia researchers: SubCell discovered three types of characteristic cellular microtubule localization patterns in (non-ciliated) U2OS cells that relate to cilia proteins. You find a short paragraph on this observation at the end of the preprint 😃.

5/ Finally, we construct the first multiscale map of cellular proteome architecture directly learned from images, obtaining associations beyond what is possible with human eyes—such as proteome complexes—and distinguish dynamic and stable behaviors within cellular compartments.

Check out more interesting new ciliary proteins in the preprint, such as metabolic enzymes, a new cilia marker protein (CFAP300), or CREB3 in cilia, for which we found a coding genetic variant related to blindness and skeletal changes!.

Our atlas portrays the cilium as a dynamic sensory organelle, suggesting that individual cells can fine-tune their cilium - both in terms of receptors and signaling components - to sense and integrate diverse signals, computing a response to relay to the cell body.

70% of cilia proteins cell-type-specifically localize to primary cilia, 78% show variation across cilia in the same cell type. In 90% of the images they localize elsewhere in the cell - at the same time (multilocalization). A great example: AKT3.

We reveal many signaling proteins in cilia, also of downstream signaling cascades, portraying the cilium not only as an antenna but also for downstream cellular computations.

Manually annotating all generated data, we revealed 654 proteins at cilia with their subciliary localization pattern – distinguishing primary cilium, tip, transition zone, and basal body localizations.

Over 2.5 years, we had a microscope running day and night - recording 17,092 confocal 3D image stacks of antibody stainings for 1,935 proteins in three diverse human cell lines, containing (according to CiliaQ) in total 463,368 cells with 218,412 cilia.
