For those in the TL;DR set, the team found that: 1. PARP14 can label a minimal 5-mer peptide; 2. It prefers a basic-K/R-E-X-X motif in vitro; 3. MAR esters on D/E are way less stable than we thought; and 4. Glycohydrolases might be sensitive to proximal sequences too!. 2 of 4.

I cannot recommend the Cohen lab highly enough as a place to compete a postdoc! If you are looking for a dynamic, inclusive lab that values big swings, you should really consider applying. Plus, the PNW is a great place to live!.

A big shoutout to the Fuller lab @SantaClaraUniv for helping us make peptides for this work. We are happy to share any details on how to adapt this technique in your lab and we hope it helps you learn more about the PARP family!. (4/4).

They demonstrate that this technique works with both peptides and proteins, using it to find a new PARP14 selective substrate peptide. And, it can be used with chemical treatments to determine acceptor site chemistries. (3/4).

Our team of enterprising undergrads - led by co-authors Sean Wallace and Leila Chihab - used a neat MALDI trick to analyze multi-site ADP-ribosylation in the closely related PARP14 and PARP15 enzymes. (2/4)
