This has been (and continues to be) such a fun project, due in no small part to the amazing undergraduate and master's student's I've gotten to work with (and @Daugherty_Lab too).

Most of what we know about immunity comes from humans & mice, but there’s LOTS of evolutionary diversity in immune proteins. We show how this diversity translates to functional diversity for a single function of RIP kinases, but clearly there’s lots more to explore 6/.

We found that diverse RIPK3 proteins activate NF-kB, including RIPK3 from species that do not have complete necroptosis. These data suggest that NF-kB activation is the ancestral & conserved function of RIPK3, and it has been co-opted to activate necroptosis in tetrapods. 5/.

RIPK3 is amongst the most rapidly evolving of the RIP kinases, and is most often associated with necroptotic cell death. It’s known that many vertebrate lineages, including birds, marsupials, and carnivores, lack a complete necroptosis pathway. 4/.

We found that diverse metazoan RIPK1 and RIPK3 proteins activate NF-kB, although there are some species-specific requirements for activation, including differential dependence on endogenous RIPK1 and/or the RHIM. 3/.

Interestingly, the core interaction motif (RHIM) of RIPK1, RIPK3, ZBP1, and TRIF is incredibly well conserved. We propose that the central function of this domain is in activation of NF-kB. 2/.
