We applied the method to study ZNF598, an E3 ligase that detects collided ribosomes. We found that ZNF598 can detect ribosomes beyond the leading one in the queue. It is a limiting factor in ribosome associated quality control.

We used very-fast CRISP to induce single DSB and observed transcription repression in minutes. The repression propagates along chromosome for hundreds of kb. Interestingly, the rapid repression doesn't depend on H2AK119 ubiquitination, but relies on proteasome to remove RNAPolII.

Interestingly, when imagining with high laser power or treating cells with oxidative stress, mRNAs are still recruited to PBody, but don't decay. This suggests that the function of PBody may be context dependent.

We observed mRNAs induced to decay were recruited to PBody and rapidly disappear, presumably decayed there. With modeling, we showed that the decay is faster inside PBody. Alternative models requiring no decay or decaying with the same speed did not fit the data.
