LFN-Acr/PA is the first protein-based anti-CRISPR delivery system and the most potent known cell-permeable CRISPR-Cas9 inhibitor, significantly improving the utility of CRISPR for genome editing. 7/9.

PA-mediated delivery of LFN-Acr reduced the off-target activity of CRISPR systems and increased genome-editing specificity. 6/9

We used LFN-Acr/PA to inhibit Cas9-mediated knockout, knock-in, transcriptional activation, and base editing in human cell lines, stem cells, and 3D cell cultures. 5/9

PA delivers LFN-Acr into cells in the same way it delivers LF. However, unlike LF, LFN-Acr is a nontoxic protein that binds to and inhibits the Cas9–gRNA complex, preventing DNA cleavage. 4/9.

PA is a pore-forming protein that anthrax toxin uses to deliver the cytotoxic protein lethal factor (LF) into mammalian cells. LFN-Acr is an engineered fusion protein composed of the nontoxic N-terminal domain of LF (LFN) and a phage-derived anti-CRISPR protein (Acr). 3/9.

The “antidote” is LFN-Acr/PA, a two-component protein system derived from anthrax toxin. 2/9
