Using public short & long-read RNA-seq data (thanks @GTExPortal and @ENCODE_NIH), we see that mRNAs using upstream TSSs tend to use upstream PASs (downstream sits are similarly paired). (3/8)

There are lots of alternative 5' transcription start (TSS) and 3' polyadenylation (PAS) sites, but, unlike for splicing, little is known about relationships between decisions at both ends of mRNAs. (2/8)

RNA processing (ie. splicing and 3' end cleavage) is co-transcriptional and real-time kinetic competition dictates alt splicing as mRNA is transcribed. But, current models (including our own) focus only on individual steps, rather than jointly modeling these processes. 2/4
