Will SMURF1 inhibitors treat PAH ? Can we now inhibit other glycine hinge proteins ? Time will tell.

So if your target is HECT E3, this worked for us. If your target has a glycine hinge, might be worth thinking about ?.

. and then ran a computational screen identifying inhibitors that function through the same allosteric mechanism.

So Gali and Amir threaded the amino acid sequence of another HECT E3, E6AP, into the structure of inhibited SMURF1.

But they were right, turns out the flexible amino acid at the elbow of the hinge is the same in all HECT E3’s, across animal, plant, and fungal kingdoms.

So if you know how that works, you should be able to drug similar proteins… now that sounds like reviewer 3😉 That’s unfair – the reviewers were helpful and really improved the research, even if it took almost two years from first submission to publication🌟.
