Proliferating cells did not have a sufficient amount of ferrous iron to break the cage, therefore, TRX-CBI conjugate remains stable and hence non-toxic. 6/6.

We then designed a ferrous iron-activatable drug conjugate TRX-CBI. When SCs exposed to TRX-CBI, the high level of ferrous iron undergoes a Fenton reaction and breaks the cage to release the cytotoxin CBI within the cell which leads to the selective killing of SCs. 5/6.

First, we tested if ferroptosis inducer drugs can be used as a senolytic and found that FIN56 selectively ablates both types of SCs. Due to the limitations of using ferroptosis inducers as a senolytic drug in the clinic, we look for alternative approaches. 4/6.

To find a common senolytic target for primary and paracrine SCs, we run an RNASeq analysis and found ferroptosis and iron metabolism-related genes as top hits. We found that SCs accumulate high levels of iron, specifically the toxic form of iron called ferrous iron. 3/6.
