These findings highlight a novel adaptation shared between the Alveolata, by which transcription factors evolved into RNA-binding proteins. Targeting this unique mitoribosome assembly machinery could open new possibilities to treat diseases such as malaria and toxoplasmosis. ๐Ÿฆ ๐Ÿ’Š.

Surprisingly, these AP2s (mtAP2s) donโ€™t bind to DNA but ribosomal RNA! Cryo-EM showed T. gondii's mitoribosome are highly complex, with over 120 proteins and more than 40 fragmented RNA pieces. โ„๏ธ๐Ÿ”ฌ The mtAP2s stitch all these pieces together, creating a functional mitoribosome.

Alveolates have the smallest mitochondrial genome! It only encodes for 2-3 proteins and highly fragmented ribosomal RNAs ๐Ÿงฌ Besides, 4 transcription factors (ApiAP2s) were found in T. gondii as essential components of its mitoribosomes โšก๏ธ๐Ÿฆ .

In this article, a repertoire of apical polar ring proteins was studied and assigned to the different APR layers. Among them, APR2 contributed to parasite's motility and invasion and its depletion caused destabilisation of the APR middle layer and F-actin leakage to the cytosol.
