Our session explored how studying bioenergetics enzymes via structural biology in divergent protist sheds light on the role of these enzymes and their assemblies across eukaryotes and on how the function in the context of each organisms’ specialised needs.

Other complex III inhibitors are being developed as drug leads for apicomplexan diseases. Together with Stone Doggett and colleagues at @OHSUNewswe we showed how the drug-lead ELQ-300 binds Toxoplasma complex III and designed a more potent version.

Atovaquone is a clinically used drug for apicomplexan diseases such as #malaria and #toxoplasmosis, but its precise mechanism of inhibition was unknown. We showed how atovaquone interacts with Toxoplasma complex III and why it is selective to apicomplexan compared to human.

@AndrewEMaclean Respiratory complexes form “super-complexes” whose physiological function is debated, and assembly only studied in a few organisms. We showed that apicomplexan conserved proteins mediate III2-IV supercomplex formation, and that this provides competitive growth advantage.
