XEC exhibits increased infectivity, enhanced nAb evasion, distinct antigenicity, reduced cell-cell fusion, and decreased S1 shedding, changes attributed to F59S and T22N mutations. Removal of glycosylation sites in XEC and KP.3.1.1 restored nAb titers, fusion and S1 shedding.

We observed decreased cell-cell fusion conferred by DelS31. Structurally, it strengthens the.interaction between NTD and RBD, leading the RBD to energetically favor the down conformation, thus becoming less accessible to ACE2 and some class 1 and class 4 antibodies.
