Reducing Derlin activity and/or inducing ER stress could potentially be a way to reduce the effect of Notch(gf)- dependent overproliferation/tumours.💊🚑👩‍⚕️ 5/6.

However reduction of Derlin activity did not strongly enhance a Notch(loss-of-function) phenotype. It didn't suppress overproliferation in general either! We think this suppression applies to certain Notch(gf) alleles, specifically. 🎯 4/6.

We think the Unfolded Protein Response(#UPR) triggered by loss of Derlins, and in conditions of ER stress, serves as a protective mechanism to correct Notch(gf) overproliferation, since reducing the activity of the UPR, reversed this suppression. 🧯💪3/6.

Reduction in Derlin activity reduced the increased spread of SYGL-1, a read out for #Notchsignalling in a Notch(gain-of-function) background. Similar suppression was also seen when we induced ER stress by chemicals.✋🛑 2/6.

The bulk of my PhD work is out! Characterizing a suppressor of a strong Notch-dependent over-proliferation phenotype (aka tumour) lead us to uncover the role of ER-associated degradation proteins (#Derlin) and the Unfolded Protein Response. #ERAD #UPR #ERstress #stemcells.
