CT-inspired two-photon lets us record glutamate activity from hundreds of synapses at a time, at >1000 Hz. Really happy to share this, and there's more to come!.

Rui Liu (Kleinfeld lab) imaged the very distinct feedforward and recurrent input patterns that come into Layer 4 of mouse cortex (400 um deep) using adaptive optics 2P imaging, showing the utility of iGluSnFR3 for input-output circuit cracking.

@abhi_aggarwal1 screened millions of variants in bacteria. 1000 variants went thru @janeliagenie screen. Abhi found 15 (!) mutations that improve performance in multiple assays. Interestingly, most of them are at the interface between the two domains of the indicator.

This project started with us realizing that the specificity of iGluSnFR for synaptic release (vs spillover) is limited by its saturating activation rate; Figure from Helassa et al 2018. This damps responses close to release sites. Biophysical modeling told us: we can fix that!

Why did we make a new glutamate indicator? Neural computation hinges on how neurons transform inputs into outputs. We can record spiking output with electrodes/Ca2+ imaging, but recording a neuron's inputs is much harder. Neurotransmitter imaging could solve this!.
