We discovered that iNKT cell-neutrophil coinfiltration on the contrary correlates with poor clinical outcomes highlighting the importance of iNKT cells in the pathophysiology of CRC.

Our findings identified a pro-tumorigenic role for iNKT cells in CRC and a functional crosstalk with tumor-associated neutrophils in murine models but is it relevant for patients? Several studies described a favourable prognostic significance for neutrophil infiltration in CRC.

Then, we started wondering if we could change functions of iNKT cells from pro- to anti-tumor even in the immunosuppressive CRC microenvironment. Turns out that adoptive cell therapy with activated iNKT cells in MC38 CRC reduced tumor burden and neutrophils infiltration.

We discovered that actually iNKT cells are protumoringenic in CRC and promote the recruitment of neutrophils with PMN-MDSCs-like phenotype and functions.

Interesting but we still did not know if iNKT cells were really protumorigenic in CRC. We used the AOM-DSS and MC38 CRC mouse models in mice lacking iNKT cells.

How iNKT cells acquired this tumor-associated phenotype? Turns out that the #oncomicrobe Fusobacterium nucleatum induces IL17 and GM-CSF expression in iNKT without affecting their cytotoxic capability but promoting iNKT-mediated recruitment of neutrophils through IL8 production.

We enrolled 118 CRC patients and we discovered that iNKT cells are enriched in tumor lesions. Tumor-infiltrating iNKT cells expressed both IL17 and GM-CSF, a unique feature compared to other conventional and unconventional T cells and correlated with the abundance of TANs.

iNKT cells are considered an attractive platform for cancer immunotherapy. However, their functional role in CRC is still controversial, limiting their potential therapeutic use.
