Taken together, this work shows that aggregation of synuclein causes physical damage to the nuclear membrane, which results in nuclear dysfunction, and that this is a toxic gain of function of pathological synuclein.

This altered nuclear morphology is associated with functional deficits in the nucleus including mislocalized RNA, altered nuclear envelope repair, and increased sensitivity to environmental toxins.

In both mouse and human patients, these nuclear and peri nuclear aggregates alter neuronal morphology, and fracture the inner most layer of the nuclear membrane, the nuclear lamina.

We show that synuclein aggregates in cortical neurons form impressive ultrastructures around the nucleus in the PFF model of synucleinopathies. These are very similar to patients with LBD.
