PGS partitioning analysis highlights a major contribution of evolutionary constrained regions to prediction accuracy and the largest per-SNP contribution from non-synonymous SNPs.

Findings reveal a significant interaction between the use of annotation data and SNP density, suggesting potential benefits of using sequence variants with annotations for prediction.

It significantly improves prediction accuracy across complex traits and common diseases in both European and non-European populations. Annotation data provide comparable information to the use of another GWAS dataset of the target ancestry.
