@isemevol @nanopore @illumina MSMC2 analyses show a slowly steadily declining trend, and we could see the separation into 2 pop! It probably happened after the last glaciation. Yet demographic (DILS) analyses infer some possible geneflow between the 2 pop, suggesting they do not deserve a species status. 6/7

@isemevol @nanopore @illumina But we first wanted to make sure that MSMC2 was performing ok in the range of parameter of this low mutation rate insect, as it could have a huge impact in SMC analyses! Simulations show that giving a good prior was helping the to MSMC model to recover the correct trend. 5/7

@isemevol @nanopore @illumina Is this low genomic diversity a consequence of a declining trend of the species? We had to look at the demography of the population, for example with SMC methods. 4/7

@isemevol We collected and sequenced in @nanopore long-reads + @illumina short-reads (polishing) 4 individuals, and looked at mito and nuclear diversity. It is very low! But O.alexandrae seems not to be an exception as some related species were also low in diversity (Troidini). 3/7

@isemevol This huge butterfly has a restrained distribution in PNG and is separated in 2 populations. Are these populations divergent and/or belonging to the same species? This question could have an impact on its conservation… 2/7
