And promiscuous deubiquitinases (DUBs) can exhibit linkage-dependent activities in the presence of UBQLN2-polyUb condensates (e.g., K63-Ub4 is protected from DUB activity - black curves below)

Importantly, UBQLN2 condensates with K63-polyubiquitinated substrates would protect these substrates from proteasomal degradation even when proteasomes are recruited into condensates. This behavior depended on the efficiency of UBQLN2 oligomerization (wild-type vs. dSTI1-II)

Consistent with prior results (, we found that K63-polyubiquitinated substrates sedimented and phase separated with UBQLN2 more efficiently than K48; but both polyubiquitinated substrates can drive UBQLN2 phase separation at very low uM concentrations.
