What was remarkable was that varicosities remain quiescent if they are not juxtaposed against dopamine receptor expressing processes.

We show that dopaminergic varicosities are very selective in how they release dopamine. They synapse onto processes that express dopamine receptors to release dopamine.

The released dopamine is understood to signal diffusively, in a rather imprecise and inefficient process that is often referred to as volume transmission. This study basically calls that model into question.

This simple observation is inconsistent with how we are taught to think of neuromodulatory release sites like that of dopamine. Our current understanding holds that release occurs stochastically from varicosities that do not form synapses.

We recently posted a preprint from my lab, summarizing work over the past couple of years where we show that the neuromodulator dopamine is released from varicosities that synapse onto the processes of target neurons.
