The robust and Ca2+ activity is visible using a GCaMP6 variant that is tethered to the outer surface of the ER. So much is happening there. Can’t wait to find out what it does.

We also found that Ca2+ waves originate from the ER and are SOCE, Ryanodine receptor and Gap junction dependent. Genetic interference of these components leads to severe stimuli-induced seizures, supporting the physiological relevance of these Ca2+ waves to brain excitability.

By performing detailed Ca2+ imaging studies, we show that perineurial cells exhibit robust, complex and dynamic Ca2+ activity that significantly differs between perineurial cells that occupy different brain regions.

We found that perineurial knockdown of dStim, acting in the Store-Operated Calcium Entry pathway (SOCE) leads to severe seizures.
