@AshimaBhakri @AnjanaRaoLab @mliarispe @StephanieEisen @basulabcolumbia Lastly, as a new PI, I had to overcome a lot of “self-inflicted (self-)doubt” every single day. I never expected this to be the first paper from our lab. I guess science plays out in its own mysterious ways and my two cents are to remember that you are not doing this alone!! 10/n.

Since several subunits of different BAF complexes are frequently mutated in cancers, the inflammatory signatures uncovered by our studies here may be relevant to pathogenesis of cancers with BAF loss-of-function mutations. Surely more to come, as we follow up on this work 8/n.

Our studies add to an emerging paradigm, where unrestrained inflammation appears to dictate the quality of the adaptive immune response. Such a feedback mechanism might be useful to prevent the breakdown of immune tolerance and autoimmunity in highly inflamed tissues 7/n.

Importantly, dampening of the inflammatory signals either by IL1b blockade or treatment with Dexamethasone, an anti-inflammatory glucocorticoid receptor agonist, led to a partial but clear rescue of germinal center responses in Arid1a-deficient mice 6/n.

A fascinating vignette: the inflammatory programs turned on in absence of Arid1a is associated with loose nucleosomal packing near transcription start sites, suggestive of changes in nucleosomal positioning potentially driven by altered histone modifications 5/n.

A surprising finding was a strong induction of inflammatory signals in B cells lacking Arid1a. Unlike the well-known functions of BAFs as positive regulators of gene expression, this suggested a role of Arid1a-BAF complex in suppressing inflammatory gene programs 4/n.

Similar to known functions of BAF in creating open chromatin landscapes, we noted that the Arid1a-BAF complex collaborates with several transcription factors of ETS, EBF and E2A family to establish chromatin accessibility at thousands of regions during B cell activation 3/n.

In this work we investigated the role of the canonical BAF complex (a.k.a. SWI/SNF @swisnf) in generation of humoral immune responses. We discovered that the Arid1a-dependent BAF complex is indispensable for production of high-quality antibodies in the germinal centers 2/n
