++.Data gaps, interpretability, & handling complex protein structures remain open questions. We highlight integrating multimodal data (e.g. sequence, structure, knowledge graphs, gene expression, text, etc.) and dynamic/temporal modelling as the next big leaps. .

++.We explore small-molecule targets and protein-based drugs (e.g. antibodies). From repurposing existing compounds to designing new therapies, pLM-based methods are boosting speed and accuracy in the drug discovery pipeline.

++.We break down two main strategies: (1) End-to-end models that learn directly from data and (2) Models leveraging huge pre-trained pLMs like ProtBERT or ESM. Each offers unique benefits and challenges for drug–target predictions.

++.In our survey/analysis study, we reviewed a nearly comprehensive list of pLM-based drug–target Interaction (DTI) prediction studies and highlighted each one according to original contribution(s) to the fast-growing literature.

++.Protein Language Models (pLMs) decode protein sequences much like how NLP models interpret text—revealing hidden structural & functional insights. Our review discusses how these pLMs have the potential to revolutionise early-stage drug/lead discovery.
