Fear #engrams are very popular but what about #SpatialMemory engrams? Here is what we learned about #cFos, #GranuleCells and navigation in freely swimming mice. Check out our paper on @NatureComms carried out at the @OertnerLab 🧵(1/5)

Here is where it gets tricky! These “engram cells” do not re-express cFos although they are electrically active! We used #CaMPARI2 to show how WM-tagged cFos cells fire (electrically active) specifically in the WM and yet they rarely express cFos. (4/5)

Using #optogenetics, we found that silencing cFos+ cells from the first WM-training day impairs memory retrieval in novice and expert mice. This suggests that indeed cFos+ granule cells are the “WM engram”, or at least part of it (3/5)

cFos re-expression (encoding vs. retrieval, aka. “reactivation”) is a criterion to define an #EngramCell. We found that cFos expression patterns in the #DentateGyrus change dramatically from day to day in a water maze spatial learning paradigm, regardless of training level (2/5)

So excited to be able to present in person again. Please join me for a discussion on how cFos temporal instability might contribute to pattern separation of memories. Today at #FENS2022 (Sun. July 10th at 14:00). #Engram #Optogenetics #SpatialMemory
