Finally, we show SMAC binds to BIRC6 via multi-site mechanisms, resulting in incredibly tight binding. This enables SMAC to competitively displace caspases and antagonise BIRC6 anti-caspase function. (5/7)

We reveal BIRC6 regulates cell death by ubiquitinating caspases-3, -7 and -9 and directly restricts caspases-3 and -7. Showing for the first time the direct inhibition of caspases by BIRC6. (4/7)

Using in vitro biochemical assays, we show BIRC6 is an unusual E2/E3 hybrid ligase working exclusively with the non-canonical E1 enzyme UBA6 to ubiquitinate its substrates, including its regulator SMAC: (3/7)

Our cryo-EM structures of BIRC6 alone and in complex with SMAC reveal BIRC6 is an anti-parallel dimer and surprisingly looks like a crab! The flexible N-terminal substrate-binding modules act as claws guiding potential substrates toward a central mouth-like cavity. 🦀 (2/7).
