Our findings reveal that variants decreasing hydrophobicity in the core of titin domains cause DCM, an information that can not only inform pathogenicity assessment of new variants but also point to new therapeutic strategies.

In this work we examined the deleterious features of pathogenic variants in titin, a key protein of cardiomyocytes that has been extensively linked to disease. We have focused on variant C3575S, which doesn't alter protein structure but significantly compromises protein stability.

DCM is a common inherited heart disease and a frequent cause for heart transplants. Recent advances allow enhanced care for cases with an identified genetic cause, but distinguishing pathogenic variants from benign polymorphisms remains a challenge.

Our paper on #titin missense variants causing #DCM is out in @CellReports! 🎉 Combining #XrayCrystallography, #Biochemistry, #MolecularDynamics, and #Genetics, we found that variants decreasing hydrophobicity in the core of titin domains cause #DCM.
