Further work is required to understand how cell context or disease status influence the mutagenic potential of NHC. The paper is open access and primary datasets are available.

What did we do? We quantified DNA damage by sequencing DNA from cell lines treated with NHC. Inactivating uridine-cytidine kinase 2 (Uck2) lowered the toxicity and mutagenicity of NHC. Limits? We only tested cell lines, so there is still lots to learn about NHC in normal tissues.

We found that in cell lines pyrimidine salvage stimulates the mutagenic activity of the antiviral NHC, the active component of molnupiravir. This metabolic quirk may explain why NHC induces high levels of DNA damage in cells grown in the lab, but not in safety tests in animals.
