Tau lesions in chronic traumatic encephalopathy (CTE) are also sites of concentrated glial reactivity. In our recent paper we used multiplex immunohistochemistry to explore the microenvironment of these lesions.

We investigated markers of microglial and astrocyte reactivity, different tau species, ubiquitination, neurofilaments, neurons and blood vessels in lesion and non-lesion areas of frontal cortex from CTE cases.

It's been a brilliant week at #AWCBR. Surrounded by the best people in one of the most beautiful places in the world. Also very grateful to give one of the plenary talks, and have such a substantial time slot for our CTE neuropathology symposium. 1/3

We show that tangles can be classified into different maturity levels using antibody labelling signatures. This reproducible and efficient method of quantifying the maturity of individual tangles will be useful for future studies of tau pathology.

We co-labelled four antibodies associated with early and advanced tangle maturity and used @FijiSc to segment individual tangle objects. Using spectral clustering we identified five distinct populations of tangles in Alzheimer's disease brains.

We were intrigued by @DrChristinaMo and @DrNeuroChic excellent 2021 review on neurofibrillary tangle maturity and wondered if we could apply our multiplex IHC approach to identify an immunophenotype signature for tangle maturity.
