If the scan step is equal to the pitch of the #SPADarray, each image is shifted by the right amount needed to sample different points of the sample. Thus, the images can be fused together, enabling an upsampled reconstruction with half the original pixel size. 🧵(4/6)

The benefits of s²ISM extend to any additional dimension attached to the ISM dataset, such as time and spectrum. We demonstrate superior fluorescence lifetime imaging, made possible by the timing capabilities of the #SPADarray and made more robust thanks to our approach. 🧵5/7

Data acquired with a #SPADarray are almost exclusively affected by photon shot noise. Thus, we maximize the Poisson likelihood to fuse the scanned images of the ISM dataset into a single super-resolution image. We call this method multi-image deconvolution. 🧵(2/6)

The idea behind s²ISM is simple: light distributes differently on the #SPADarray, depending on the axial position of the emitter. With proper statistical modelling of ISM image formation, we reconstruct only the in-focus fraction of the collected fluorescence photons. 🧵2/7
