Here we describe a molecule (METTL7A) acts in the pre-implantation period to attenuate oxidative stress that enhances embryo development to the blastocyst stage and subsequent conceptus elongation in cattle.

The bXENs and blastoids established here represent valuable cell models to study embryogenesis and the pregnancy loss. Upon further test their developmental potential, bovine blastoids will help develop new ARTs for improving reproductive efficiency in livestock species (6/n).

Finally, we developed an improved bovine blastoid using bXENs. We showed that this 3L-blastoids have a similar proportion of hypoblasts and a slightly higher ratio of epiblasts compared to day8 blastocysts, with majority hypoblasts surrounding epiblasts (5/n).

Furthermore, by treating bovine IVF embryos with bXENs signaling cocktail, we showed that the growth and progression of epiblasts are also facilitated in the developing pre-implantation embryo (4/n).

We implemented a 3D co-culture model with bXENs to examine whether or how hypoblast regulates the development of epiblast during bovine embryogenesis. We demonstrated that the presence of bXENs and associated communications support the growth and stemness of bEPSCs (3/n).

We conducted comprehensive molecular characterization of bXENs. Transcriptomic annd epigenomic analyses confirmed the identity of bXENs and revealed that they are resemble hypoblast lineages of early bovine peri-implantation embryos (2/n).
