             Lastly we induced inflammation and found vasculature released cytokines for potential immune cell infiltration (IL-1β, IL-6, IL-8) and polarization (GM-CSF, IL-12p70) which was absent in avascular tissues. This underscores the importance of integrating vasculature for IBD studies

             We then incorporated a microvascular network interacting with the crypts. HUVEC and fibroblasts self-assembled to form blood vessels at the bottom of fibrin gels and were ~70 µm from crypts. This close proximity suggests potential crosstalk between epithelium and vasculature

             We found proliferative cells (red) could spontaneously localize within the crypts compared to planar tissues as well as crypt-patterned tissues without basolateral flow. The results illustrate the significance of both crypts and basolateral flow for in vitro colon models.

             A short thread 🧵 highlighting our key findings, We developed a method to pattern fibrin hydrogels with a crypt topography and populate the crypts with colon cells of ~600 µm depth and observed minimal gel remodelling.
