This raises the possibility that autosomal genes in other cell types with a dispersed XIST configuration (pre-implantation embryos, germ cells, immune, lung, and cancer cells) may be regulated by XIST. (5/6).

We made similar discoveries in differentiating mouse PSCs where Xist is in a dispersed configuration and autosomal localization also occurs, suggesting that dispersed configuration of XIST/Xist is coupled with autosomal spreading. (4/6).

Applying RAP-seq, we found that XIST spreads across the X and, surprisingly, also to autosomal genes. Autosomal targets show female-specific and XIST-dependent H3K27me3 and downregulation of gene expression, indicating that XIST enforces sex differences on autosomes. (3/6).

In female naïve human pluripotent stem cells (PSCs), XIST is dispersed in the nucleus and X-chromosome dampening (XCD) occurs, raising the question of whether XIST mediates XCD. Knocking out XIST, we found that it is required for XCD in naïve hPSCs. (2/6).

In collaboration with @dinodicarlo, we used hydrogel nanovials to capture and measure the secretion of VEGF-A alongside mRNA transcriptomes. Surprisingly, we found that the VEGFA transcript was not the best predictor of secretion! 2/2.
