Mechanistically, we define a U6-complementarity rule that predicts QKI dependence, establishing a U6-anchored splice-fidelity checkpoint at weak 5'SS. QKI binds donor-proximal introns, directly engages U6, and, with U4/U6·U5 tri-snRNP, secures exon inclusion
