Could tolerance to DNA be broken in the gut in systemic lupus erythematosus?. In our latest review we explore whether tolerance to DNA can be broken when undigested DNA from microbiota in GALT directly encounters B cells unrestricted by T cell dependence.

Normal functions of DN2 B cells, DNASE1L3 and C1q in the gut likely relate to bacterial recognition, bacterial killing and disposal of bacterial debris including DNA. Failure of this system could result in persistence of bacterial DNA that could stimulate an autoimmune response.

By a combination of spatial transcriptomics and multiplexed single-cell technologies, DN2 B cells (previously known to be expanded in blood in lupus) were found to interact in GALT with dendritic cells co-expressing lupus autoantigens DNASE1L3 and C1q, adjacent to bacteria.
