The efficiency of vector-based CRISPR is low compared to RNPs in Ulva, but we show that co-editing of APT with a non-marker gene is feasible. We managed to install targeted mutations in three additional genes.Additional optimisations will be necessary for multiplex CRISPR.

We evaluated vector-based CRISPR. We tested 31 vector configurations and found that bicistronic fusion of Cas9 to a resistance marker or incorporation of introns in Cas9 led to the most mutants. Curiously, vector DNA was often inserted at the target site.

We observed that delivery of two RNPs resulted in the creation of large deletions in the APT marker gene. @juliarraiza helped to consolidate this observation and we could readily produce 20 kb deletions of Genomic DNA around the APT locus.

Efficient CRISPR-based genome editing is difficult to achieve in many (macro)algae. After some initial tests, we benchmarked CRISPR editing of the marker gene APT using RNPs and a non-marker gene using targeted insertion of a resistance marker.

We describe a chemical sensitivity screen for Ulva and its mutualistic bacteria. Ulva is sensitive to Blasticidin and we can confer resistance via Blasticidin deaminases. This nicely complements the current single selection systems to generate stable transformants in Ulva.
