Out now in @PLOSGenetics! Activation of ChvG-ChvI regulon by cell wall stress confers resistance to β-lactam antibiotics and initiates surface spreading in Agrobacterium tumefaciens. It's great to see my first co-first author paper published!.

Summary of our conclusions: ChvG/ChvI has a conserved role in sensing cell wall stress and conferring resistance to β-lactam antibiotics. Check out our preprint to see what happens if we deplete PBP1a even longer😉. Hint: ABC transporters. (10/10)

This response appears to be conserved in Sinorhizobium meliloti and check out preprint of Alex out of the @HallezLab to see similar results in Caulobacter crescentus! (9/10).

Two-component systems (TCS) are often used by bacteria to rapidly assess and respond to environmental changes. The ChvG/ChvI (ChvGI) TCS conserved in α-proteobacteria is known for regulating expres...

So does ChvG/ChvI respond to cell wall stress? We treated chvG and chvI deletion mutants with β-lactam antibiotics to find out. Turns out they were extremely sensitive to β-lactams, but not any other class of antibiotic we tested! (8/10).

And finally, deletion of exoA, a gene required for the biosynthesis of succinoglycan, restored microcolony formation! (7/10)

Next we looked at succinoglycan production. Succinoglycan is an exopolysaccharide secreted by Agrobacterium and other plant-associated members of the Rhizobiales. Succinoglycan pooling was observed in the PBP1a depletion using calcofluor staining. (6/10)

So then what causes the spreading? We explored major operons that were differentially expressed in the depletion: flagellar motility, type VI secretion, and succinoglycan biosynthesis. We found that the spreading was independent of flagellar motility and type VI secretion! (5/10)
