CSF1R signalling sustains glucose uptake in Ly6Chigh monocytes to fuel the hexosamine biosynthetic pathway (HBP), which in turn allows optimal CSF1R expression. Impairing glucose flux towards HBP through GFPT1 invalidation leads to increased Ly6Chigh/Ly6Clow ratio.

We report that surface CSF1R expression relies on continuous translation and export to the membrane. CSF1R recycling rate accelerates as monocytes differentiate from the Ly6Chigh to the Ly6Clow subset, and CSF1R blockade prevents Ly6Chigh commitment towards this trajectory.

We report that this subset is conserved across several tissues and especially fat, in which they regulate lipid metabolism. Importantly, these cells have a rapid turnover from monocytes, and in particular MDP-derived monocytes that are biased to differentiate into CD226+ Macs !
