The same effect occurred when pulling bead 2 while observing bead 1. But when the beads were separated by tens of microns, the force-transmission signature disappeared 📉, showing that force propagation diminishes with distance.

Here’s the interesting part: a smaller stress-relaxation signature also appeared in bead 2’s force data 📈, even though no direct manipulation was applied! This signature reveals mechanical force propagation across the axon membrane.

When bead 1 was pulled, the force-time data showed a stress-relaxation curve—a sharp force peak followed by a relaxation phase. This reflects tether formation mechanics: tension rises as the tether stretches and drops as more membrane material flows in.

We observed a neuron and used two fluorescent beads (let’s call them bead 1 and bead 2) to extrude tethers from the axon while tracking force and position for both beads.
