I’m thrilled to share our preprint presenting MORSE: a new way to simultaneously and quantitatively track patterns of neuromodulators/neuropeptides in vitro & in vivo using spatially multiplexed 3D imaging of 10+ green optical sensors on a microendoscope.

10/ Therapeutic implications? Targeting NKCC1 or SPAK might mitigate injury progression in infants, while interventions like MMP inhibition (PMID: 38190820) could address damage in toddlers. Age-specific therapies could be key!.

9/ Human autopsy data corroborated our findings, showing region-specific alterations in pNKCC1 after pediatric TBI, particularly in damaged white matter & the ChP. The parallels between swine & human data are striking.

8/ TBI also disrupted chloride cotransporters in the choroid plexus, with pNKCC1 signal becoming dysregulated. This suggests ionic homeostasis at brain-fluid interfaces is critical post-injury.

7/ We next employed IHC for neuronal pNKCC1, and found it significantly increased in infant piglets with TBI, and across both ages, amount of neuronal pNKCC1 positively correlated with seizure duration and injury severity.

6/ Interestingly, TBI affected the cortex & hippocampus differently. In our model, the cortex exhibits structural damage, but the hippocampus is spared. Yet, in both regions, infant piglets with TBI showed increased NKCC1, KCC2, and SPAK compared to toddlers.

5/ Key finding: In swine, the GABA switch occurs during infancy across cortex and hippocampus, aligning with data in humans. This makes swine an excellent translational model for pediatric brain injury research.
