Together, our work shows that heterogeneous progenitor contributions to tissue architecture-defining cell type proportions and postembryonic organ growth are key mechanisms forming the adult liver.

Interestingly, we observed a transient appearance of polyploid cells during a massive postembryonic growth phase.

Extending lineage tracing to adulthood determined that embryonic cells undergo spatially heterogeneous three-dimensional growth linked to distinct environments. Strikingly, giant clusters comprising almost half a ventral lobe suggest lobe-specific dominant-like growth behaviors.

Moving to the in vivo system we probed this prediction with lineage tracing experiments using the multispectral FRaeppli system and identified not only unipotent hepatoblasts, but….

1. Mathematical modeling predicts that a heterogeneous progenitor lineage potential is sufficient to achieve 1:9 BEC-to-hepatocyte ratio proportions.

1. First, we analyze cell type proportions of differentiated hepatocytes and biliary epithelial cells (BECs) in vivo. We show that a precise 1:9 BEC-to-hepatocyte ratio is established fast. But how is such a ratio established?

How do liver progenitors contribute to the functional organ? Happy to share this work, in which we used lineage tracing in the zebrafish 🐟 liver to study the contribution of individual liver progenitors to the different cell lineages and postembryonic organ growth🧵👇 (1/10).

To meet the physiological demands of the body, organs need to establish a functional tissue architecture and adequate size as the embryo develops to adulthood. In the liver, uni- and bipotent...
