Biological conduction bridges skip a beat. The preprint authors generate AV node-like pacemaker cells from human pluripotent stem cell populations in order to create a biological conduction bridge.

CD34 specifically labels SAN pacemaker cardiomyocytes in human SAN tissue and SANLPC cultures. Sorting for CD34+ cells from human pluripotent stem cell differentiation cultures enriches for SANLPCs with a functional pacemaker phenotype.

We show that the developing SAN consists of three subtypes of pacemaker cells: Core Pacemaker, Sinus Venosus, and Transitional Cells. Human pluripotent stem cell-derived SANLPC cultures contain the same subset of pacemaker cells and closely resemble fetal SAN pacemaker cells.

We performed single nuclei transcriptome analysis of the human SAN to get a better understanding of its cellular and molecular composition and identified multiple SAN specific markers including MYH11, BMP4 and CD34.
