These findings highlight the significant role of tissue mechanics in regulating immune responses. Understanding this could lead to novel therapeutic targets for treating chronic inflammatory conditions and preventing excessive coagulation.
We explored how mechanical forces—specifically cyclic stretch and substrate stiffness—affect NETosis. We found that increased stretch and stiffness amplify NETosis and sensitivity to NET-inducing agents via activation of Calpain, PI3K, and FAK pathways.

Shear stress increases intracellular calcium levels in neutrophils and this process is mediated by Piezo1. Activation of Piezo1 in response to shear stress mediates calpain activity and cytoskeleton remodeling, which triggers NETosis. @Ying25505110 @DrQuinn4realz @ManijehKh2020.
