So I’ve been working in the lab brushing up on protocols and techniques. I put a jellyfish gene into E. coli so they produce GFP, the same protein that makes jellyfish light up.

For 15 years, my daily routine has included weed and dabs. It's just. the way it is. But for a while now, I've started feeling like my tolerance is so high I'm not even getting high anymore, and I'm honestly starting to wonder if the habit is holding me back in some areas.

Pretty sure it’s something to do with the IPTG and induction. Colonies aren’t glowing. The Kan +dna grew, while the Kan -dna didn’t, so the controls worked. I will redo the plates and try to do a liquid induction into fresh lb + kan broth I started. Science! 🧪

I harvested over 2000 tomatoes this season from this setup with just 6 plants!

The glow doesn’t show until I add IPTG. I set that up yesterday, so by Monday I should see my first glowing plates under blue light if everything went right. It’s exciting to think there’s a jellyfish gene working inside bacteria I genetically modified myself! 🧬

I tried two ways to get the DNA inside. The chemical method (calcium + heat) didn’t really work, but the electric pulse method did. That little shock opened the cells and let the plasmid in. The ones that survived carry the new gene and can live on antibiotic plates.
