We performed a 16h timelapse of triple-negative breast cancer spheroids undergoing collagen-invasion, not only characterizing the presence and properties of invading phenotypes but also finding a way to figure out whether a treatment is specifically targeting them or not! 9/11

We performed a 24-hour timelapse of M1 polarization of macrophages with a gene expression reporter, finding that polarization-specific phenotypes are indeed overexpressing a polarization-specific gene! We now have a direct connection to explore with other omics techniques! 7/11

What about time? Here's a 23-hour timelapse of keratinocytes undergoing calcium-induced differentiation! We show not only massive phenotypic changes but also notice the cells organizing in patches while differentiating, showing the importance of the spatiotemporal context! 5/11

We show that cell lines have different organelle phenotypes, but what about finer differences like stress response?. We exposed our MCF10A cells to a number of stressor showing different mechanisms depending on the stressor as well as unique, stress-dependent phenotypes! 4/11

Combining hyperspectral imaging, environment-sensitive organelle dyes, phasors, bioimage and data analysis, we created ESPRESSO (Environmental Sensor Phenotyping RElayed by Subcellular Structures and Organelles) that can track cell states over 24 hours continuously! 3/11

High-dimensional phenotyping with transcriptomics or proteomics are "snapshots" in time, how can we obtain a similar wealth of information, but time-resolved? . Lots of papers hint at organelle properties and their link to cell state, can it be something to exploit? 2/11.
