Some Ribo-Seq datasets show lower similarity with experimental profiles. With these datasets we also find other indicators of poor dataset quality - projecting your datasets into polysome profiles is a good tool for checking dataset integrity.

Combined Ribo-Seq and RNA-Seq datasets can be projected into modelled polysome profiles. Many published datasets yield similar modelled and experimental profiles. The two very different methods yield consistent information about cellular protein synthesis.

Polysome profiles (a method for assessing cell-wide translational activity) have a predictable mathematical structure. Any method that reveals ribosome densities can be used to simulate polysome profiles (yellow line) and compare them to experimental profiles (black dots).
