🔑 Key Findings:.* LIG1 loss is synthetically lethal with PARP inhibition. * LIG1 is altered in 6% of CRPC patients. * Combined LIG1 and PARP inhibition triggers replication stress and DNA damage, ultimately leading to apoptosis. * LIG1-PARP synthetic lethality is conserved.
