While the leaf disks plasmolysed I prepared the whiskers. I gently shook the tubes manually for 30sec every 3 mins for 15mins. To the second tube I omitted plasmid DNA; neg control for RUBY but positive control for whiskers. 6/10

I then placed the remaining disks, 6 to each tube in 1mL of plasmolysis buffer in sterile round bottom tubes. The buffer will shrink the inside of the plant cells to make a cavity between cell wall and cell membrane. This helps plant brace itself for wounding. Incu RT 30mins 5/10

Then I punched out 18 leaf disks. Plated the first 6 on MS with hormones as a positive control for regeneration. These will be whisker free and non transgenic. 4/10

Next I surface sterilized the leaf using NaDCC bleach and 0.01% triton as surfactant for 20mins followed by two rinses in sterile distilled water. I prepared a plasmolysis buffer in the meantime. Mannitol 5g MES 100mg Distilled Water to 50mL pH adjust to 5.7 with 1M KOH 3/10

My UVC mutant snapdragon flowered and its beautiful!!! It has that clamshell phenotype that Ive grown fond of and seems to have very little if any yellow hue. So lovely. My first unique bloom 🥰💚

And this is when I spin the tubes down. The glow in the leftmost tube fades to a dull background while tube 2 and 3 remain glowing, indicating no binding. Tube 4 is neg control; no DNA just whiskers. I think this worked?(!)
