We found Ven-sensitive myeloma exhibit low heme biosynthesis and increased hemin uptake. Heme promoted Ven resistance via activation of RAS-RAF-MEK signaling and purine metabolism. Targeting heme biosynthesis or co-targeting BCL-2/MCL-1 overcomes heme-induced Ven resistance (2/3).
