Hi Brian. 37C works fine as well. We decided to go with 30C to be able to compare curing efficiencies between pFREE and the temperature-sensitive version pFREE-RK2. And also to reduce activity of Cas9 a bit when it cures ON.
@norholm_m @lauritsen_i Hi Morton and Ida, sorry 1 more question! Methods say that you kept strains and did the curing at 30*C, is there a specific reason? ie if I did everything at 37*C does that make a difference to curing efficiency?
